Pinky, the tiny fish, lives in the sea with his small friends. The big fish make fun of them.

Would it be correct if I removed "The" from the second sentence. It would then read: Big fish make fun of them.

It would be correct, but the meaning would be slightly different. Using the definite article 'the' means that some (specified) big fish make fun of Pinky and his friends. You would needed to identify or refer to these - e.g. Pinky the tiny fish lives in the ocean [etc] with other fish of various sizes. The big fish make fun of the small ones.

Omitting the definite article means that some (unspecified) big fish (e.g. ones who happen to swim by) make fun.
