I and my girlfriend want to have a "happy time" in a room of certain hotel or inn.
Is there a word to contain the whole procedure?
I mean that once the word be mentioned,the counterpart will get the meaning.
Another term might be "check-in." When you arrive at the hotel, you "check-in" at the front desk or office to get your room assignment, the keys to the room, and present your method of payment.

would you say"come on! honey let's check-in,let's tryst,let's registration"

I think you're looking for one word for the whole affair. It may not exist, a quickie is implied in the waggle of an eye or rendering of a cliche.
A tryst is correct, but it doesn't necessarily mean to check in or register unless you do so under the name Mr. and Mrs. Smith and pay with cash.

In conversation, you might talk of "getting a room".
To the hotel staff, you might talk of "privacy".
To your girlfriend, you probably won't have to say anything at all. Ladies are smarter.
Anyway, the hotel just rent the room. It is entirely up to the guests what they plan to use it for.

There is no single word in English for what you're wanting to express. The phrase that is closest to what you want to express to your girlfriend is "let's get a room." She'll understand what's implied.
