It's a little shop of my own.
It's my own little shop.

Is the first sentence correct? If so, what is the difference in meaning between the two sentences?

Yes. So is the second. They mean pretty much the same thing. First one may emphasize the shop a bit more, second emphasizes you a bit more, but the difference between them is really minimal.
