What are the differences in" despite", "in spite of","in despite of",and "despite of"

No, there is really no difference in those expressions. Don't let the word "of" confuse you, it is a preposition, and is required grammatically. The expression "in despite of" is very uncommon, and i would recommend that you do not use it.
