To express the meaning "Have you understood my point?", which of the following is acceptable?

A. "Understand?"
B. "Understood?"

Both work fine, but I'll venture that B sounds like it's more apt to be used when a speaker seeks to be more strident.

Do you understand? vs Is that understood?

I'd use, "Understand?" when talking about factual matters to an intelligent equal.

I'd use, "Understood?" for an unruly inferior.

B. Have you understood that?

It's hard to say definitively, ILQ, because the delivery, the intonation, would make a difference in the intended meaning. While one collocation will tend towards strident, it doesn't mean that it will be so in all situations.

I have to echo JTT.

Conversational English depends on who is saying what to whom in what context as well as on formal grammar.

i'd say 'is that understood'
and 'do you understand that',
because, i dunno, maybe it's just me, but have you understood that doesn't sound as good as is that understood
