Could I use YES to replace NO without change of meaning?

WB that's another really good q. Myself I might long said, "Oh Yes, no...," depending of course on how one situate quote marks
Luna misspoke. She was answering the question she thought she heard, rather that what was actually asked.
But that's quite common in everyday dialogue.
