If I were to title a book 'The Word Ambidextrous', would the grammar be okay? Ought it to be 'The Word 'Ambidextrous''? The latter looks right, but it looks bad in a book title. Any ideas?

I think it would be okay. You don't have to worry too much about grammar in a title anyway. Some titles are deliberately awkward so as to be eye-catching.

How about

Ambidextrous: The Word? Maybe not.
