Does it mean finding new possibilities?

Both "knocking on doors" and "turning over stones" have the same general meaning here -- dilligent search and research.

Yes. Unless you are looking for insects, the last place you would look for something is under a stone. If you are turning over stones, you are looking just everywhere.
