LOL! Arthur - the joke is the play on the word "come".

It can - and is intended by the first nun - to mean "I have never travelled using this route before".

It can also mean to orgasm - which is the sense which the second nun assumes to be the case.

The cobblestones are relevant because they could, theoretically, provide a vibration sensation - which could cause a woman to achieve orgasm, when ridden over on a bicycle.
