In most eukaryotic cells, subsets of microtubules are adapted for specific functions by post-translational modifications (PTMs) of tubulin subunits. Acetylation of the epsilon-amino group of K40 on alpha-tubulin is a conserved PTM on the luminal side of microtubules that was discovered in the flagella of Chlamydomonas reinhardtii.

The luminal side or surface of something enclosed such a cell or tubular organ is the surface facing inwards.

Acetylation of the epsilon-amino group of K40 on alpha-tubulin is a conserved PTM on the luminal side (inward facing surface) of microtubules. It was discovered in the flagella of Chlamydomonas reinhardtii.

This is rather heavy-duty scientific writing. It appears to be about the virus that causes the sexually transmitted disease, chlamydia.

A flagella is a hair. Teeny tiny organisms use them for propulsion. Luminal refers to the lumen, the interior of a hollow structure. It's an inner membrane space per Wikipedia.

In this case the sentence "Acetylation of the epsilon-amino group of K40 on alpha-tubulin is a conserved PTM on the luminal side of microtubules that was discovered in the flagella of Chlamydomonas reinhardtii. " seems unacceptable in the sense of usual English grammar.
