The two phrases have similar meanings and can sometimes be used inerchangeably, but also they can have slightly different meanings -

What do you think about something - please give me your extended opinion of the topic - e.g. if you ask me what I think about the war in Afghanistan, I might tell you my opinion of whether my country's forces should be involved, their chances of success, what I think might happen in the future, what I hope will happen, etc. What do you think of something - please give a brief answer indicating approval or disapproval. If you ask me what I think of your new hat, I might just say "very nice".
