Is 'more' required?

...in which a more rigorous and [more?] challenging workload is offered.

Would I have to insert the 'more' before 'challenging' in the sentence above? Which is favored in formal writing?

Either is correct, so more would not be required. Sounds like something someone would put into a job resume, though. The use of 'more' indicates the old job was also challenging, but the person wants to indicate he is looking for more challenge than in the last position. If that's the context, I would leave it in.

Using the word "more" once in front of "rigorous and challenging" implies it is modifying both words.

If you didn't want to modify the word "challenging" with "more", you would place it before, as in ... challenging and more rigorous.
