It is a school whose teachers had give up (hope) on their students.

Is the sentence correct? Is 'hope' necessary?

no, hope is not used correctly.
either of these are correct:

It is a school whose teachers had given up on their students.
It is a school whose teachers had given up hope because of their students.

Region Philbis is right.

Please note, to 'give up' on something is rather different in meaning from to 'give up hope'.
