(Not 'on' or 'in')

That sentence needs attention! Firstly the repetition of 'petrol' is poor style. You could write "He filled up at the petrol station", the mention of the petrol station being sufficient to indicate that fuel was being purchased.
If you need to specify that it was petrol and not diesel or LPG or hydrogen that was being bought, you could write "He filled up with petrol at the
service station"

No need for 'some', and it would be very foolish and dangerous to fill the car itself with petrol.
To "fill up on" is what people do when they eat something.
The children would not eat their supper. They were not hungry as they had filled up on potato chips during the afternoon.
