My professor says that my sentence " How do you differentiate between people who are introverts and extroverts" is wrong. She says it should be "How do you differentiate people who are".... the use of 'between' is incorrect. Is this true?

To differentiate is to distinguish between, which is possibly what you meant. In the phrase "differentiate between" the word "between" is redundant. Your professor is correct.
