If you go to www.m-w.com (our usual source) and type in "gladlier", you will go to the listing for "glad" where "gladly" is listed. The way that website works, if they didn't accept "gladlier" as a word, they would say "The word you've entered isn't in the dictionary". So, apparently it's a word! It works the same way for plurals, etc.
