Should I say/write, "the car's front seat" or "the front seat of the car"?

Is it "the door's handle" or "the door handle"?

Either is fine but it would depend on the sentence as to which sounds better. Often you would only use "the front seat" because "of the car" would be obvious from the context of the conversation.
