Dr. Marco Vietor and Paul Crusius have been creating successful businesses since college.

Should it be "since they were in college"?

In this case, "college" refers to a time period so it's fine as it is. This is a common usage although I doubt you would find it in a dictionary. You might see this construction anytime there is a key life event. "Since birth", "since bootcamp", "since high school".
