The idiom "signed off" is usually accompanied by "on", as in, "He signed off on the order".

Is the comma after "in" required?

No, it is not. Were I to use any punctuation in such a situation, I'd use a colon, thus ' . . . as in, "He signed off on the order".' No punctuation is required, however.
