I'm not sure that I agree.

It looks to me as if "enduring and overcoming all the difficulties" belongs together as a phrase.

It is possible that a hyphen might be used instead of a comma though. Although I think the comma probably works.

your perseverance, endurance and conquest over all the difficulties along the way.

It is the embodiment of both your earnest faith desiring a temple in your country, and your perseverance -- enduring and overcoming all the difficulties along the way.
