Hello.
When we are talking about the timing of the school day, should I say, e.g.

1. I have biology in the 6th period.
or
2. I have biology in 6th period.

An American would most likely say "in sixth period." I don't know if the term "period" referring to the divisions of the school day is used by other native speakers of English.

i recall people saying it without the preposition.

"i have biology sixth period" or "i have sixth period biology"...

Yes, I've heard that, too. The significant thing for this question is that I've never heard anyone use the definite article.
