When writing a letter, which should I use, #1 or #2? If neither, what should it be?

How are you using them, exactly? Why are they grouped?

The hyphens are used if it's a part of an official title, e.g. Detachment Officer-in-Charge, Assistant Officer-in-Charge, Officer-in-Charge of Aircraft Intermediate Maintenance Detachment Sigonella.

There is no single rule or custom governing hyphenation of job titles. It is necessary to ascertain the appropriate local custom.
