William Sydney Porter, who wrote under the name of O. Henry, was a United States short-story writer, well known for his descriptions of the many kinds of people he observed and knew in New York City.

Is the comma needed after 'writer'?

It's not necessary because the clause after the word "writer" is a modifier proceeding the independent clause.
