I had the depo on the 13th February for the first time I have been experiencing spotting but this only occurred in the last three days im supposed to go and have the jab again tomorrow I have been sexually active with my partner so I was wondering
A) Could the depo jab have failed
B) How long after having only the one jab would it take for it to be out of my system as we want to have a baby together

You need to talk to the doctor who gave you the depo injection. Ask him or her! This is why they are there. And if they blow you off, find another doctor. You are a consumer of medical services just as much as you are a consumer of coffee or manicures or whatever. Vote with your wallet and reject any doctor who will not answer basic questions on the medical care that they have given to you.
