Can you request a relationship with someone who already is in one on Facebook or will it just say pending until they confirm?

You request to be "FRIENDS" with someone - not a relationship.
Yes, that person must accept you as a friend.

What if you are already friends with this person? If they accept the relationship request, will it automatically end with the person they were "in a relationship " before?

As I said in your other, nearly identical topic - try blocking this person and I suspect the Facebook database will figure out that you are no longer in a relationship together.
