Are they natural English? Is the indefinite article optional? Are there other ways of expressing the same idea?

The second one is grammatically correct but I'm not quite sure what you want to ask...

Are you room mates?
Are you in a relationship with your room mate?
Are you sleeping with your room mate?

Is S optional? PRESENTATION here means a student/students working on a subject/given topic and then reading aloud the handout/powerpoint file in class.
